Cutting hole in new bathroom vanity countertop...

Should I go ahead and mount it to vanity and THEN MEASURE AND CUT HOLE?

OR should I cut hole before mounting counter to vanity?
 
Mount it to the vanity first (even as a dry fit if necessary). Sometimes the fit isn't quite right and you have to sand the sides/edges. ALSO, make sure your cabinet frame does not get in the way of the new sink/faucet set. I have seen ambitious sinks that were too large ... and took up all the counter room with nothing left for the faucet!

Check your margins carefully! :D

The countertop is set.
The sink is rectangular, so I should be good on room.
